MOTS-c Usage and Storage Guide
MOTS-c mitochondrial-derived peptides:
MOTS-c is a peptide derived from mitochondrial DNA and is the first mitochondrial-derived peptide discovered. As a key metabolic regulator, it plays a central role in energy homeostasis, insulin sensitivity, and metabolic adaptation, especially in responding to metabolic stress. Unlike conventional peptide hormones, MOTS-c is directly encoded by mitochondrial genes, and its unique nucleus-mitochondrial communication mechanism provides a novel perspective for studying metabolic diseases such as aging, exercise adaptation, and type 2 diabetes. Its ability to promote glucose uptake and enhance metabolic flexibility has made it a focus of attention in the field of metabolic research.
Because MOTS-c can mimic some of the benefits of exercise and directly activate the AMPK pathway, it is favored by many researchers focused on metabolic health and aging. It does not act through traditional hormone receptors, but rather by regulating cellular metabolic state, avoiding complex downstream signaling interference. Its core functions lie in optimizing mitochondrial function, promoting glucose utilization, and enhancing antioxidant defense, making it a unique and powerful experimental tool for studying age-related metabolic decline, exercise physiology, and insulin resistance mechanisms.
Applications of MOTS-c mitochondrial-derived peptides:
MOTS-c optimizes mitochondrial function, promotes cellular glucose uptake, enhances insulin sensitivity, and improves the body's adaptability to metabolic stress. It is a regulatory peptide based on mitochondrial-nuclear signaling and plays a crucial role in maintaining energy homeostasis.
MOTS-c has been used in experiments requiring precise regulation of energy metabolism, mimicking the benefits of exercise, or combating age-related metabolic decline. Its mechanism of action bypasses the classic insulin signaling pathway, directly improving cellular metabolic efficiency and stress resistance. Therefore, MOTS-c is a highly promising research molecule in exploring the molecular basis of metabolic syndrome, sarcopenia, the biology of aging, and exercise intervention.
MOTS-c Usage and Storage Methods
1. Reconstitution and preparation :
Recommended solvent :
Preferred solvent : sterile water for injection or phosphate-buffered saline with pH 7.2-7.4
Stability optimization : Sterile PBS containing 0.1% bovine serum albumin
For cell experiments : Dissolve in sterile water and then dilute with serum-free culture medium.
Standard preparation procedure :
Solvent preparation :
Solvents were sterile filtered through a 0.22 μm filter membrane.
Return the solvent to room temperature (20-25°C) to avoid overheating.
Reconstitution procedure :
Add 1.0 mL of the recommended solvent to a 10 mg vial.
Pour slowly along the bottle wall to avoid vigorous foaming.
Gently rotate the vial for 2-3 minutes, let it stand for a while, until the lyophilized powder is completely dissolved and the solution is clear.
Final concentration: 10 mg/mL (approximately 4.6 mM)
Dispensing and dilution :
Immediately aliquot into 20-50 μL portions using low-adsorption centrifuge tubes.
Dilute to the working concentration using the appropriate buffer (such as PBS or culture medium) according to experimental requirements.
Clearly label the name, concentration, preparation date, and batch number.
Working concentration reference :
In vitro cell experiments : The commonly used working concentration range is 10 nM to 1 μM
In vivo animal studies : The commonly used dosage range is 0.1 to 5 mg/kg/day.
Example of preparation calculation : If a 1 μM working solution is required, take 0.22 μL of stock solution and add it to 1 mL of culture medium.
Key points to note :
Handle gently : Avoid swirling or violent shaking to prevent protein denaturation.
Aseptic operation : Performed in a laminar flow hood, using sterile pipettes and tubing.
Strictly avoid light : It is sensitive to light, and all operations and storage must be carried out under light-protected conditions.
Use immediately after reconstitution, or complete dilution and dispensing within 1 hour.
Avoid repeated freeze-thaw cycles : Diluted working solutions are not recommended for long-term storage and should be prepared and used immediately.
2. Storage conditions :
Unreconstituted lyophilized powder :
Long-term storage : Store frozen at -20℃, shelf life 24 months.
Optimal storage : Store frozen at -80℃, shelf life 36 months.
Keep away from light : Must be stored in the original light-proof aluminum foil bag.
Moisture protection : Keep dry. Contains desiccant. Seal after opening.
Reconstituted solution :
Use immediately : For best activity, use immediately after reconstitution.
Short-term storage : Store at 2-8℃ away from light for no more than 12 hours.
Repackage and freeze : Store at -20℃ or -80℃ away from light for no more than 72 hours.
Repeated freeze-thaw cycles are prohibited : a maximum of one freeze-thaw cycle is allowed, otherwise the activity will be significantly reduced.
Transportation conditions :
Freeze-dried powder : transported by dry ice (-78℃).
Short-term shipping : Transported with ice packs at 2-8℃, and delivery is guaranteed within 24 hours.
Packaging : Light-proof aluminum foil bag + vacuum seal + foam insulated box.
3. Stability and Processing Standards :
Chemical and physical stability :
It is relatively stable in solutions with a pH of 6.5-7.5.
Avoid contact with organic solvents, strong acids, strong alkalis, oxidants, and detergents.
Avoid high temperatures, violent vibrations, and ultrasonic treatments.
Key points for preserving active ingredients :
Proper dispensing and storage at -80°C are key to maintaining long-term activity.
Before use, slowly thaw the dispensed solution on ice to avoid repeated freeze-thaw cycles.
