1. Reconstitution and preparation :
Recommended solvent :
Preferred solvent : Dimethyl sulfoxide (DMSO) . This product has good solubility in DMSO and can be prepared into high-concentration stock solutions (e.g., 50-100 mM).
Aqueous solvents : Since 5-amino-1MQ is usually present in the form of salts (such as iodide, toluenesulfonate), it also has some solubility in sterile deionized water or phosphate-buffered saline (PBS) , but may require slight heating (such as a 37°C water bath) and brief vortexing to aid dissolution.
Precautions : DMSO is hygroscopic and must be sealed after opening. High concentrations of DMSO are toxic to cells; the recommended final working concentration for in vitro cell experiments is no more than 0.1% (v/v) of DMSO .
Standard preparation process :
Calculate the required stock solution concentration. For example, to prepare a 50 mM (50 mmol/L) stock solution:
The molecular weight is 159.21 g/mol.
Weigh or take 5 mg of this product; its amount of substance is approximately 5 mg / 159.21 mg/mmol ≈ 0.0314 mmol.
To obtain a concentration of 50 mM, the required solvent volume is 0.0314 mmol / 0.050 mol/L = 0.628 mL.
Therefore, adding approximately 0.63 mL of DMSO to a 5 mg sample yields a stock solution of approximately 50 mM.
After adding the calculated solvent, gently vortex or let stand at room temperature for several minutes until the powder is completely dissolved and a clear solution is formed.
Dilute the stock solution to the working concentration using the appropriate cell culture medium or buffer (ensuring that the DMSO is adequately diluted) as required by the experiment.
Working concentration reference :
In vitro cell experiments : The commonly used working concentration range is 1 μM to 100 μM . The specific concentration needs to be optimized according to cell type, exposure time, and research objectives (such as IC50 determination). Dose-dependent experiments are recommended.
In vivo animal studies : Dosage needs to be determined based on the animal model, route of administration (intraperitoneal injection, oral gavage, etc.), and experimental design. Doses in mouse models reported in the literature can be used as a reference, but must be validated independently .

The preparation process should be carried out in a clean environment to avoid microbial contamination.
