Progesterone (progesterone) chemical properties and uses explained

Progesterone is a naturally occurring progestogen with the chemical name pregnene-4-ene-3,20-dione , a molecular formula of C₂₁H₃₀O₂ , and a molecular weight of 314.46 g/mol . It is a key hormone in the female menstrual cycle, pregnancy maintenance, and embryonic development . It is also secreted in small amounts in men (by the adrenal glands and testicles).

1. Chemical properties of progesterone

Appearance : White or off-white crystalline powder.

Solubility :

It is insoluble in water but easily soluble in ethanol, acetone and chloroform .

Clinical preparations are mostly oily injections, vaginal suppositories or oral micronized preparations .

Melting point : about 121-125°C .

Stability : Sensitive to light and heat, need to be stored away from light.

Steroid skeleton :

Contains 4 rings (A/B/C/D) , with a keto group (C=O) at position 3 and a double bond at position 4 .

Biosynthesis :

It is converted from cholesterol to pregnenolone and then to progesterone (dependent on 3β-hydroxysteroid dehydrogenase ).

Liver metabolism : converted to pregnenolone and pregnendiol (excreted in urine).

Half-life :

Oral administration : About 5-20 minutes (strong first-pass effect, requiring special preparations to improve bioavailability).

Injection : Approximately 18-36 hours (oil sustained release).
